From formulation release mechanisms to pharmacopoeia compliance: Compatibility assessment of drug formulation characteristics with USP Apparatus 1–7 dissolution test apparatus.

From formulation release mechanisms to pharmacopoeia compliance: USP dissolution testing apparatus (Apparatus 1-7) selection evaluation and suitability analysis

In formulation R&D, analytical method validation, and batch quality control (QC release), the precise application of Apparatus 1 to 7 devices, based on drug release mechanisms (immediate release IR, sustained release ER/CR, enteric-coated delayed release DR), physical specific gravity (floating/sinking), and solubility characteristics (BCS classification), to correspond to the United States Pharmacopeia (USP) <711> Dissolution and USP <724> Drug Release, is a crucial foundation for establishing stable IVIVC (in vitro-in vivo correlation) and compliance with international inspection regulations (TFDA, FDA, EMA, PMDA).

I. USP Apparatus 1: Rotating Basket

  • Core mechanism: 40-mesh stainless steel swivel basket (optional 10/20/100-mesh or PTFE/gold-plated acid-resistant basket), speed typically 50-100 RPM.

  • Most suitable drugs and dosage forms:

  • Capsules: soft and hard gelatin capsules, plant-based capsules (avoid floating on the surface of the liquid).

  • Low-density/floating tablets: such as gastric retention sustained-release tablets and porous foaming granules.

  • Controlled-release microparticles (Pellets, Beads): Placed in a microporous mesh basket for dissolution.

  • Hydrophilic Suppositories.

  • Selection limitations: If the dissolved gelatin adheres to the mesh, it can easily cause clogging, resulting in an artificially low dissolution rate.

  • [Chengwei Technology's Distributor Models]:

  • Pharma Test's flagship dissolution analyzer series from Germany: PTWS 130D/S (6-slot), PTWS 820D / PTWS 830 (8-slot QC standard model), PTWS 1420 (14-slot high-throughput model) – all models are equipped with the patented MonoShaft quick-release system, allowing tool-free 3-second basket switching, and strictly controlled axial runout (Wobble) to < 0.2 mm.

  • Logan Instruments Automated Dissolution Series: UDT-818-8 / 850DLA (8 slots), UDT-860-12 / 860DLA (12 slots ICH M9 dual-zone speed control), UDT-818-18 / 880DLA (18 slots flagship verification model) – two-piece quick-release drive shaft, highly automatic positioning, supports USP 1 automated sampling.

II. USP Apparatus 2: Paddle Method

  • Core mechanism: PTFE-coated or 316 stainless steel stirring paddle, bottom clearance 25 ± 2 mm, speed typically 50–75 RPM.

  • Most suitable drugs and dosage forms:

  • Immediate-Release (IR) Tablets: Global >70% of oral solid dosage forms meet the gold standard for QC release.

  • Conventional oral extended-release tablets (ER tablets): matrix tablets and hydrophilic colloidal coated tablets.

  • Oral suspensions and emulsions.

  • Chewable Tablets.

  • Selection limitations: Samples that are prone to floating should be used with sinkers; heavy, insoluble excipients are prone to forming a "coning effect" in the center of the cup bottom, and can be evaluated with a peak vessel (permitted by regulations).

  • [Chengwei Technology's Distributor Models]:

  • Pharma Test series from Germany: PTWS 820D / PTWS 830 (8-slot golden ratio) and PTWS 1420 (14-slot double row), equipped with a micro-vibration independent external water bath circulating heater (37.0 ± 0.2°C), meeting the latest FDA mechanical validation (MQ) standards.

  • Logan Instruments (USA) series: UDT-818A-8, UDT-860-12 (12-slot independent dual-speed control, capable of 6 vs 6 parallel comparisons of original/generic drugs), UDT-818-15 / 870DLA, UDT-818-18 / 880DLA (CDMO process validation three-batch simultaneous 6×3 slots).

III. USP Apparatus 3: Reciprocating Cylinder (BioDis)

  • Core mechanism: A flat-bottomed inner cylinder (5-40 DPM) is embedded inside the glass outer tube and reciprocates up and down. Multiple rows of outer tubes automatically move laterally to switch between different pH media.

  • Most suitable drugs and dosage forms:

  • pH-dependent enteric-coated formulations (Delayed-Release / Enteric-Coated): Enteric-coated microcapsules, enteric-coated tablets (automatically simulating a complete release trajectory at gastric acid pH 1.2, duodenal pH 4.5, and ileal pH 6.8).

  • Colon-targeted drug delivery: for those who need to undergo multiple changes in the intestinal physiological environment over a long period of time.

  • Beads & Multi-particulates: The reciprocating tumbling action can realistically simulate the mechanical shearing force of gastrointestinal peristalsis.

  • Selection limitations: Not suitable for immediate-release tablets; requires high maintenance of mechanical consumables and multi-row pipelines.

  • [Chengwei Technology's Distributor Models]:

  • Logan Instruments BioDis Reciprocating Cylinder Dissolution System – Equipped with a patented automatic flip-top anti-evaporation mechanism and a tapered condensation reflux tank (seamless coverage during transverse runout, completely eliminating evaporation loss and cross-contamination), driven by a brushless DC motor from 4 to 40 DPM (accuracy ±0.1 DPM), supports 7 test tubes + 1 isothermal solvent refill tube, and can be integrated with the DSC-800 intelligent automatic sampling workstation.

IV. USP Apparatus 4: Flow-Through Cell Method

  • Core mechanism: A columnar flow cell (12 mm or 22.6 mm) is paired with a pulseless precision dual-piston pusher, supporting both open-loop and closed-loop modes.

  • Most suitable drugs and dosage forms:

  • Poorly soluble drugs (BCS Class II & IV): Open-loop continuous flow maintains an infinite sink condition, eliminating the need for large amounts of surfactants that compromise physiological realism.

  • Long-acting microsphere injections (Microspheres / LAI): such as PLGA sustained-release microspheres (long-acting monitoring for weeks to months).

  • Nano suspensions and liposomes: paired with a dedicated ultrafine dialysis membrane flow cell.

  • Subcutaneous/muscle implants and drug-eluting stents (DES).

  • Lipid-soluble suppositories: They melt and float at 37°C, but can still be continuously wetted and desorbed by a stable fluid in a flow cell.

  • API Powders: Determination of the intrinsic dissolution rate of trace powders.

  • Selection limitations: The flow path system requires high precision and has strict requirements for degassing and de-bubbling.

  • [Chengwei Technology's Distributor Models]:

  • Logan Instruments (USA) Series: Logan Super-4 Flow-Through Dissolution Tester – Supports both open-loop and closed-loop modes, equipped with 8 modular quick-connect flow cells (compatible with 22.6mm, 12mm, powder, suppositories, stents, and dialysis clamps), dual-piston pulseless push flow (0–24 mL/min, volumetric accuracy 0.1 mL), built-in MC-4000 four-channel pH gradient automatic switching system, and can be coupled with online UV-Vis/HPLC/Permetro biomimetic osmosis cells.

V. USP Apparatus 5: Paddle over Disk Method

  • Core mechanism: Place a dedicated stainless steel mesh tray into the bottom of the standard Apparatus 2 tank, fix the patch flat with the adhesive side facing up, and keep the stirring paddle 25 ± 2 mm away from the mesh surface.

  • Most suitable drugs and dosage forms:

  • Transdermal Patches (TDS): Standard-sized local analgesic patches, nitroglycerin, nicotine, and hormone patches.

  • Buccal & Sublingual Patches.

  • Selection limitations: It can only evaluate the dynamics of patch drug release into the medium and cannot simulate the permeation resistance of the human skin stratum corneum (percutaneous penetration test requires the use of Franz Cell / FTC-800 flow cell).

  • [Chengwei Technology's Distributor Models]:

  • Pharma Test series from Germany: PTWS 820D / 830 / 1420 with original USP Apparatus 5 disc fixture kit - compliant with USP <724> precision geometric clearance specifications.

  • Logan Instruments UDT-818-8 / UDT-860-12 series from the USA, with a dedicated Paddle-over-Disk mounting disk.

VI. USP Apparatus 6: Rotating Cylinder Method

  • Core mechanism: A stainless steel cylindrical rotating drum replaces the stirring paddle, with a patch flatly attached to the outer wall of the cylinder, immersed in 1000 mL of medium and rotated at a medium speed (25-50 RPM).

  • Most suitable drugs and dosage forms:

  • Large Surface Area Transdermal Patches: A standard alternative when the patch is too large to be applied flat to the Apparatus 5 disc.

  • Polymeric Transdermal Films.

  • Selection restrictions: Applicators must be highly skilled to prevent edge detachment or microbubbles.

  • [Chengwei Technology's Distributor Models]:

  • Pharma Test series from Germany: PTWS 820D / 830 / 1420 with MonoShaft USP Apparatus 6 dedicated rotary module – mirror-polished corrosion-resistant rotary drum, fast coaxial locking, completely eliminating rotary drum sway.

  • Logan Instruments (USA) Series: UDT Series Dedicated Rotating Cylinder Drive Components.

7. USP Apparatus 7: Reciprocating Holder

  • Core mechanism: The clamping fixture moves up and down with short amplitude (standard 2 cm) in a small volume sample tube (5-50 mL), and automatically moves and rearranges the tubes at regular intervals.

  • Most suitable drugs and dosage forms:

  • Low-Dose Transdermal Systems: Trace amounts of drug in a 900 mL cup are easily below the HPLC detection limit (LOD), and the small size of the Apparatus 7 allows for significant enrichment of sample concentrations.

  • Osmotic Pump Tablets (OROS): Laser-drilled osmotic-release tablets.

  • Miniature ophthalmic/subcutaneous implants: such as intraocular pressure-lowering stents and long-acting subcutaneous hormone stents.

  • Selection restrictions: Samples require dedicated fixtures (spring clamps, mesh clamps, three-dimensional fixing frames).

  • [Chengwei Technology's Distributor Models]:

  • Logan Instruments BioDis Reciprocating System (Apparatus 7 Dedicated Configuration) – Featuring short-stroke 2 cm amplitude control, support for small-volume sampling tubes (5 mL/20 mL/50 mL) and diverse sample holders, fully automatic timed horizontal reflow, and fully compliant with FDA 21 CFR Part 11 data integrity requirements.
